摘要
目的探讨白藜芦醇对H2O2所致心肌细胞损伤的保护作用。方法采用H2O2处理培养的乳鼠心肌细胞,造成氧化应激模型,应用MTT法观察白藜芦醇对心肌细胞活力的影响,SOD及MDA试剂盒检测细胞培养液中SOD与MDA含量,并通过流式细胞仪检测乳鼠心肌细胞的凋亡情况。结果①损伤模型组与空白对照组比较,心肌细胞活力明显降低,心肌细胞凋亡率明显增加,而MDA活性明显升高,SOD活性明显降低;②不同浓度白藜芦醇可显著性提高受损心肌细胞活力,减少心肌细胞的凋亡,同时可显著降低培养液中MDA活性,而升高SOD活性。结论白藜芦醇可减轻H2O2对心肌细胞的损伤程度,对心肌细胞具有保护作用,其机制可能与其抗心肌细胞氧化和抗凋亡有关。
Objective It is to approach the protective effects of the Resveratrol on myocardial cells injury induced by H2O2.Methods Oxidative damage model were induced by H2O2,MTT was used to observe Resveratrol on the viability of cultured myocardial cells,SOD and LDH kit were used to detect the activity of SOD and LDH in cell culture medium;and flow cytometry was used to detect the apoptosis in neonatal rat cardiomyocytes.Results ① Compared with blank control group,in H2O2 model group,myocardial cell activity was significantly reduced,cell apoptosis was significantly increased,LDH activity was significantly increased,and SOD activity was significantly decreased.② Different concentrations of Resveratrol could significantly improved viability of damaged myocardial cell,reduced myocardial cell apoptosis,at the same time significantly reduced the activity of MDA in the culture medium,and increased SOD activity.Conclusion Resveratrol can mitigate injury of H2O2 on myocardial cell;it has protecting effect on myocardium cell,the mechanism may be related to its antioxidant and antiapoptosis properties.
