摘要
采用化学药物腐蚀法建立了鸡肾上腺的摘除方法,用竞争蛋白结合分析测得血浆总糖皮质激素(GCT)为38.44±9.50nm ol/L,显著低于正常鸡血浆GCT(93.50±5.78nm ol/L),并至少能维持72h,可以满足非GC依赖性生理试验的要求。给化学摘除肾上腺公鸡皮下注射GC拮抗剂RU486 缓释胶(50m g/kg),使外周血淋巴细胞糖皮质激素受体(GR)阻断率达74.93±9.59% ,而血浆GCT 维持正常水平,从而成功地建立了鸡非GC依赖性GR阻断模型。在此期间实验鸡ACTH 分泌增加,糖代谢维持正常。
The adrenalectomy in male chickens was established by the chemical corrosive method. The plasma GCT content of these chickens was cut down to 38.44±9.50nmol/l and was obviously lower than that of the normal chickens. (P<0.01).Meanhile,RU 486 ,a potent GC antagonist (in dose of 50mg/kg with polyvinyl alcohol), was subcutaneously injected in the adrenalectized chickens .Consequently the downregulation rate of glucocorticoid receptor(GR)was 74.93±9.59% in peripheral blood lymphocytes by the radioreceptor binding assay with 3H DXM;and the plasma GRT content was maintained in normal standard .The results demonstrated that GR blocked model was useful and maintained for 72 hours in the male chickens. In this period, the secretion of ACTH was increases and glucose metabolism was normal.
