摘要
采用药物缓释法,给雏鸡皮下注射糖皮质激素(GC)拮抗剂RU486(50mg/kg)阻断糖皮质激素受体,然后将雏鸡于(43±0.5)℃下实施热应激,测得其血浆GC为(113.96±16.60)nmol/L,比正常鸡约升高1倍,而外周血淋巴细胞糖皮质激素受体(GR)阻断率为41.09%±6.22%,血糖浓度为(13.89±0.60)mmol/L及血浆ACTH水平为(77.81±5.24)ng/L,与正常对照雏鸡血糖浓度及血浆ACTH水平相比无统计学差异(P>0.05);未见外周血淋巴细胞程序性死亡现象;同时观测雏鸡平均存活时间(6.29±0.31)h,为普通应激雏鸡平均存活时间的约2倍。结果表明,GR阻断可拮抗由应激导致GC过度升高对机体的不良影响,并可提高对高温的耐受性。
RU 486 , a potent glucocorticoid (GC) antagonist, (at doses of 50 mg/kg with polyvinyl alcohol) was subcutaneously injected to the chickens. The GR blocked chickens were placed in an environment of heat stress, with the temperature to be maintained at (43±0.5)℃. The experimental results were as follows: The downregulation rate of glucocorticoid receptor (GR) in peripheral blood lymphocytes was (41.09±6.22)%; the blood glucose was (13.89±0.60) nmol/L, being no significant difference between the model chickens and control chickens (P>0.05); none of the lymphocytic apoptosis to GC was found in periperal blood; and that the average survival time was (6.29±0.31) h in the GR blocked chickens, which was twice as long as that of control chickens. It was proved that RU 486 could stabilize homeostasis and elevate the adaptability to disadvantageous environmental factors.
