摘要 探讨骨形成蛋白(BMP)对人骨肉瘤细胞的作用及意义.方法:将重组人BMPZ基因的反义核酸逆转录病毒表达载体PDOR-AB用脂质体导入BMP表达阳性的骨肉瘤细胞OS-99OI,观察骨肉瘤细胞OS-99OI的BMP表达、增殖活性、肿瘤抑制基因"m23的表达、细胞周期和电镜下形态改变.结果:转染后,OS-99o1细胞中内源性的BMP表达产物明显下降;增殖活性明显下降;肿瘤转移抑制基因"m23表达未见改变;细胞周期分析表明:GI期细胞比例上升,S期比例下降;电镜观察可见细胞质中溶酶体增多,染色质有断裂现象.结论:反义核酸技术阻断BMPZ在OS-99o1细胞中的表达后,肿瘤细胞的增殖活性降低,证实骨肉瘤细胞中BMP的表达增加了肿瘤细胞的增殖活性. AIM: To further understand the role of BMP2 ex-pression in osteosarcoma cell line (OS-99O1). METHODS:This recombinant vector encording antisense cDNA of BMP2RNA (pDOR-AB) was transduced into osteosarcoma cell lineOS-99o1 by lipofectin and the biological activity of transfect-ed OS-99Ol cells was analyzed by immunohistochemistry forthe expression of introcellular BMP2, PCNA, and nm23gene, and by FCM for the morphological changes- RE-SULTS: Both the expression of BMP2 and proliferating activ-ity of transfected cells were obviously decreased and Gl phaseof transfected cells increased while S phase reduced when ob-served by FCM. There was no difference in nm23 gene ex-pression before and after transfection. The electronmi-croscopic observation showed increased lysosome chromatinchange in some cells. CONCLUSION: The results of ourstudy demonstrate that the expression of antisense BMP2RNA decreases the proliferating activity of OS-99O1 cells andBMP2 may play an important role in the biological activity ofosteosarcoma cells.
