摘要
目的:探讨He-Ne激光对TNF基因的调控作用。方法:用斑点杂交及Northern 杂交技术观测激光对肿瘤患者TNF基因表达的影响,采用24m W He-Ne激光照射荷瘤小鼠及肿瘤患者脾区(瘤区),照射前后分别取脾细胞及外周血淋巴细胞,从中提取RNA。通过双抗体夹心ELLSA 法及受体吸收试验,观测激光照射对带瘤机体TNF受体及TNF活性的影响。结果:带瘤机体TNFm RNA 及TNF活性明显降低,血清及培养上清中可溶性TNF受体(sTNFR)的表达量增高。但经激光照射后TNFm RNA 表达量显著增高,TNF活性增强,而sTNFR 含量减少,瘤块明显减小。照射前后结果比较,差异有高度显著性(P< 0.01)。结论:He-Ne激光照射具有促进TNFm RNA
Objective:To study the influence of He-Ne laser on TNF gene expression.Methods:24mW He Ne Laser was applied to irradiate spleen region with tumor in patients and mice.Spleen cell and lymphocyte in tumor body were obtained before and after irradiation, and RNA was drawn.We used spot blot hybridization and northern blot hybridization technique to observe the effect of laser on TNF gene expression in patients with tumor.Results:TNFmRNA was lower in tumor body.After laser irradiation,TNFmRNA was signifcantly raised( P <0 01),difference was notable. Conclusion:He Ne laser may promote TNFmRNA expression.
