摘要
目的:研究P物质(SP)对脾细胞免疫功能的调节作用。方法:采用3H-TdR掺入法、放射免疫测定和流式细胞仪等方法,检测了SP对体外培养的大鼠脾细胞的增殖和分泌IL-2、IL-6、TNF功能及T细胞表型表达的影响。结果:SP能明显刺激脾细胞的增殖,其中浓度为1×10- 4m m ol/L时作用最强;SP能促进T细胞CD分子的表达,使CD4+ /CD8+ 升高;SP能显著促进脾细胞分泌IL-2,对脾细胞分泌IL-6具有抑制作用,而对分泌TNF无作用。结论:证实了SP是具有广泛的免疫调节作用的一种神经肽。
Objective: To explore the effectsofsubstance P(SP) on im mune function ofspleniccells. Methods: Afterthe spleniccellsfrom ratswerecultured in vitro, theirproliferative activity was determ ined w ith incorporation of 3H-TdR, production ofcytokines detected by RIA and phenotype ofT lym phocytes investigated w ith FCM. Results: ①SPaugmented the proliferation ofsplenic cells to a peak of 1×10- 4 m m ol/L. ②SPprom oted the expression ofphenotypeofTlym phocytesto resultin theincreasein num ber ofCD+3 and CD+4 Tlym phocytes. ③SPsignificantly promotedthesecretion ofIL-2butdecreased thatofIL- 6 and did notaffectthatofTNFfrom the spleniccells. Conclusion: SPis a kind ofneuropeptide with high capacity ofimm unity regulation.
