摘要
目的:比较分泌型和跨膜型TNF-α在体内抗瘤作用的异同.方法:在肿瘤局部直接注射3种TNF-α(野生型TNF-α重组体、跨膜型及分泌型TNF-α突变体)基因,观察其抗瘤效应 结果:3种TNF-α均能有效表达。并具明显抑瘤效应,跨膜型TNF-α突变体对早期肿瘤的仰制效应明显高于其它2种TNF-α(P<0.01),而野生型TNF-α对中肿瘤的抗瘤效应则最佳.此外,注射3种TNF-α裸DNA均未观察到明显副作用,结论:分泌型和跨膜型TNF-α抑瘤机制可能下同,裸DNA直接用于细胞因子基因治疗较安全。
Objective: To study the antitumor effects of transmembrane TNF-α and secretory TNF-α in vivo. Methods: Three types of TNF-α cDNA plasmids (wild type TNF-α; transmembrane TNF-α mutant; secretory TNF-α mutant) were directly injected into tumor-tearing mice. Results: The three types of TNF-α could be expressed by tumor cells and all of them could inhibit evidently the rate of tumor growth. The tumor regression after treatment with transmembrane TNF-α mutant at the early stage was more significant than that with the other two types of TNF-α( P < 0.01) . In contrast, the antitumor effect of treatment with wild type TNF-α at the middle stage was the strongest. There was no obvious side-effect observed during the treatment. Conclusion: The mechanisms involved in tumoricidal activity of transmembrane TNF-α and secretory TNF-α may be different and the direct injection of cytokine cDNA seems to be a safe, easy and effective approach for tumor biotherapy.
