摘要
目的：研究ＴＧＦ－β及中药抗纤复方对小鼠前胶原α２（１）基因上游启动子活性的影响。方法：以含小鼠胶原α２（Ｉ）基因上游０．４、０．８．２．０ｋｂ启动片段为研究靶序列，以氯霉素乙酰基转移酶（ＣＡＴ）作报告基因，应用基因转染技术研究小鼠胶原启动子活性。结果：ＴＧＦ－β可促进小鼠前胶原α２（Ｉ）基因启动序列的启动活性，而中药抗纤复方可明显抑制其启动活性。结论：ＴＧＦ－β和中药抗纤复方都可作用于胶原α２（Ｉ）基因调控片段，进而促进或抑制胶原合成。
Objective : TO study the effect of TGF-B and Chinese herbal on promoter activity in mouse a2(I ) procollagen gene. Methods:The upper stream 0.4.0.8.2.0 kb Sequences in mouse a2(I ) procollagen gene 5' flanking region were studied with CAT as reportergene and gene transfection method Results: The result revealed that TGF-B increases promoter activity of mouse a2(I ) procollagen genewhile Chinese herbal significantly inhibits the Promoter activity. Conclusion:TGF-B Chinese herbal increase or inhibit collagen synthesis byregulating promoter element in procollagen gene.
