摘要
应用3 H—DL—谷氨酸为示踪剂进行突触小体谷氨酸摄取实验,通过体外、体内实验观察铝盐对大鼠脑突触小体谷氨酸摄取功能的影响。结果显示: 剂量为37.5~600.0μm ol/LAl3+ 在体外作用15 分钟, 能使大鼠脑组织海马区突触小体3H—DL—谷氨酸摄取抑制25.9~72.3% ;大脑皮层抑制11.0~63.4% ,且有一定的剂量-效应关系。动物以20.0、60.0m g/kg·d Al3+ 经饮水染毒50 天, 观察到高剂量时, 海马及大脑皮层谷氨酸摄取抑制了12.1% 、7.4% , 与对照比较无显著性差异 (P> 0.05)。表明体外铝对突触小体摄取灭活谷氨酸的功能有抑制作用,
Effect of AlCl 3 on glutamate uptake in rat brain synaptosome were studied with in vitro and in vivo methods.At concentrations of 150,300 and 600μmol/L in vitro,aluminum significantly inhibited 3H DL glutamate uptake in rat cerebral cortex and hippocampus synaptosome with proportions of 32 18%~72 34% and 29 75%~63 39% respectively and with a half inhibitory concentration between 150 and 300μmol/L,reflecting an obvious dose effect relationship.But uptake of 3H DL glutamate had no apparent changes after exposure to 20 and 60mg/kg·d Al 3+ in drinking water for 50 days in vivo.
