以1周龄海蓝白蛋鸡为实验对象,采用连续腹腔注射的方法建立铝中毒模型,研究铝对鸡大脑组织钙稳态的影响。实验设对照、低、中、高4个剂量组,暴露剂量分别为0、18.31、27.47、36.62mg/(kg·d)(Al3+),染铝60d后,用原子吸收分光光度法检测鸡大脑组织中铝的含量,以Fura-2/AM作为细胞内游离钙离子荧光指示剂,检测鸡大脑神经细胞中游离钙离子浓度([Ca2+]i),采用半定量RT-PCR法检测鸡大脑组织中钙调蛋白(CaM)mRNA的表达。结果表明,各染铝组鸡大脑组织铝含量、神经细胞[Ca2+]i显著高于对照组(P<0.05,P<0.01),CaMmRNA表达水平显著低于对照组(P<0.05,P<0.01);且呈剂量-效应关系。提示铝暴露可致鸡大脑神经细胞内钙稳态失调。
The model chicks was produced by continuous peritoneal injection of different concen-tration aluminium trichloride to investigate the effect of aluminum on calcium homeostasis in chicken brain at doses of Al^3+ 0,18.31,27.47 and 36,62 mg/(kg .d)for successive 60 days, The content of aluminum in brain tissues were measured by atomic absorption spectroscopy. The concentration of [Ca^2+]i in fowl nerve cells was detected by Furoa-2/AM as the calcium fluorescent indicator, Expression of CaMmRNA was detected through semi-quantitative reverse transcriptase-polymerase chain reaction(RT-PCR). The results showed that the content of AI and [Ca^2+]i in the nerve cell were higher(P〈0.05, P〈0.01)and expression of CaMmRNA was lower (P〈0.05,P〈0.01)than control significantly. The results indicated that aluminum disturbed the calcium homeostasis in the nerve cell of chicken brain.
