HIF-1α与HGF在非小细胞肺癌中的共表达及其与淋巴管生成的关系 被引量:2

Co-expression of hypoxia-inducible factor-1α and hepatocyte growth factor in non-small-cell lung cancer and their association with lymphangiogenesis

摘要 目的探讨低氧诱导因子-1α(HIF-1α)、肝细胞生长因子(HGF)在非小细胞肺癌(NSCLC)中的共表达及其与淋巴管生成的关系。方法应用免疫组织化学方法标记淋巴管,计数微淋巴管密度(LMVD),检测126例NSCLC患者及20例癌旁对照肺组织中HIF-1α、HGF的表达,分析其表达情况与LMVD及临床病理参数之间的关系。结果 HIF-1α与HGF的表达呈明显相关(R2=0.590,P<0.05);NSCLC组织中存在HIF-1α和HGF的高表达,阳性表达率分别为85.7%和80.9%,明显高于癌旁对照肺组织(P均<0.05),癌组织中的LMVD明显高于癌旁对照肺组织(17.525 0±2.286 9 vs 1.765 0±0.735 3,P<0.05);HIF-1α或HGF阳性表达的NSCLC患者较阴性者有更高的LMVD(P<0.05),HIF-1α、HGF的阳性表达与TNM分期、淋巴结转移显著相关。结论在NSCLC中存在HIF-1α与HGF的共表达,该表达可促进NSCLC淋巴管的新生及淋巴转移。 Objective To investigate co-expression of hypoxia-inducible factor-1α(HIF-1α) and hepatocyte growth factor(HGF) and their association with lymphangiogenesis in non-small-cell lung cancer(NSCLC).Methods Applying the immunohistochemical technique,the lymphatic micro-vessel density(LMVD) and expressions of HIF-1α and HGF in 126 patients with NSCLC and 20 peri-cancerous normal lung tissue samples were determined.Relationships of expressions of HIF-1α and HGF with LMVD and clinicopathological factors were analyzed.Results Expression of HIF-1α was positively related to expression of HGF(R2=0.590,P0.05).Expressions of HIF-1α and HGF in NSCLC tissues were significantly higher than in peri-cancerous normal lung tissues(17.5250±2.2869 vs 1.7650±0.7353,P0.05).Higher LMVD was found in NSCLC samples with positive HIF-1α and HGF expressions than those with negative expressions(P0.05).Expressions of HIF-1α and HGF were significantly associated with the TNM stage and lymph node metastasis(P0.05),while they had no significant correlation with sex,age,tumor size,histological type and differentiation grade(P0.05).Conclusion Co-expression of HIF-1α and HGF exists in NSCLC,moreover,over-expressions of HIF-1α and HGF can promote lymphangiogenesis and lymph node metastasis.
