摘要
[目的]为L-茶氨酸提取制备提供可靠的检测手段。[方法]采用高效液相色谱蒸发光散射检测器(HPLC-ELSD)快速定量测定白茶、碧螺春、翠竹中L-茶氨酸的含量。[结果]试验表明,在反相C18色谱柱上,以0.15%三氟乙酸(含0.06%七氟丁酸)水溶液-乙腈溶液(88∶12)作为流动相,能有效检测L-茶氨酸,L-茶氨酸浓度在0.2~1.0 mg/ml时,其峰面积(Y)与相应的浓度(X)呈线性关系,线性方程为Y=2×10-7X+0.005 7,相关系数为0.999 6。L-茶氨酸的平均回收率为99.55%~100%,相对标准偏差为0.39%~1.00%。[结论]该方法精密度、准确度好,稳定性高,能简便、快速、准确地测定茶叶中L-茶氨酸的含量。
[Objective] To provide reliable detection methods for extracting and preparing L-theanine.[Method] High Performance Liquid Chromatography-Evaporative Light Scattering Detection(HPLC-ELSD) was used to quantitatively detect the L-theanine content in white tea,Biluochun and Cuizhu.[Result] On the C18 column in reversed phase column,with 0.15% trifluoroacetic acid(containing 0.06% heptafluorobutyric acid) in aqueous solution-acetonitrile(88∶ 12) as mobile phase,L-theanine could be effectively detected.The detection limit concentrations of L-theanine was 0.2-1.0 mg/ml,the corresponding concentration(Y) and peak area(X) were linear related,the linear equation was Y=2×10-7X+0.005 7,the correlation coefficient was 0.999 6,the average recoveries of L-theanine was 99.55% to 100% and relative standard deviation was 0.39%-1.00%.[Conclusion] The method was precise,accurate,highly stable,simple,convenient and rapid for the quantitative determination of L-theanine form the extracted liquid of teas.
