摘要
建立纤维素酶辅助野马追总黄酮的最佳提取工艺。以野马追为原料,首先研究了pH、液料比、酶浓度、酶解温度和酶解时间对得率的影响。在此单因素实验基础上,优化出了纤维素酶辅助提取野马追总黄酮的最佳工艺参数:pH 5.5、液料比12:1(mL/g)、酶浓度0.4%、酶解温度40℃和酶解时间1.5h,此时总黄酮的得率为1.436%。然后将纤维素酶辅助提取法与其它提取方法进行了对比,实验结果表明:纤维素酶辅助提取法的得率比水煎煮法高96.4%,比乙醇回流提取法高37.8%,且提取时间仅为1.5h,相比乙醇回流提取法缩短了1h。该方法效率高,耗能低,省时省材料,具有较高的实用价值。
An effective technology was established for cellulase-assisted extraction(CAE)of total flavonoids from Eupatorium lindleyanum DC.Eupatorium lindleyanum DC.was used as raw material,the effects of pH value,solvent to material ratio,enzyme concentration,enzymolysis temperature and enzymolysis time on extraction yield of total flavonoids were investigated.The optimal CAE parameters of the extraction were found as follows:pH value 5.5,solvent to material ratio 12 ∶1(mL/g),enzyme concentration 0.4%,enzymolysis temperature 40 C and enzymolysis time 1.5h,and yeild was 1.436% under this extraction condition.Compared with other extraction techniques,the yeild of CAE was 96.4% higher than water decoction method and 37.8% higher than ethanol refluxing extraction.The extraction time was 1.5h and it was 1h less than ethanol refluxing extraction.This method is more efficient,saves material and time.It is a very practical method.
