摘要
目的 建立反相高效液相色谱法(RP-HPLC)测定野马追中槲皮素的含量;比较了3个产地野马追中槲皮素的含量。方法 药材经提取水解后,采用高效液相色谱法对野马追药材中槲皮素的含量进行了测定。色谱柱Kromasil-C18(4.6mm×250mm,5μm),流动相为甲醇-0.4%磷酸(50:50),检测波长360nm,流速0.6ml/min,柱温30℃。结果 槲皮素在2.78~27.8μg/ml线性关系良好(r=0.99913),平均回收率为98.698%,RSD=2.4%。结论 该方法快速简便,重现性好,为野马追及其制剂提供了一种简便准确的含量测定方法。
Objective An RP-HPLC method was established for the determination of quercetin in Eupatorium lindleyanum, to compare the content of quercetin in Eupatorium lindleyanum from different product area. Methods RP -HPLC was used to determine the content of quercetin in Eupatorium lindleyanum after herb was extracted and hydrolyzed. Quercetin was separated completely on a Komasil C18 column(4. 6 mm ×250 mm, 5 μm), using a mixture of methanol-0.4% phosphoric acid(50:50) as mobile phase. With UV detection at 360nm, and the flow rate is 0. 6 ml/min, the temperature of the column is 30℃. Results The calibration curve was linear in the range of 2.78~ 27.8 μg/ml of quercetin with correlation coefficient being 0.999 13. The average recovery was 98. 698% ( RSD = 2.4%, n = 5). Conclusion The method is quick and simple with good repeatition and can be used for quality control,
