大鼠急性癫痫发作后齿状回分子层微血管构筑的改变

Alterations of the microvascular network in molecular layer of dentate gyrus of rats with acute seizures

摘要 目的观测海人酸(KA)癫痫模型大鼠海马齿状回分子层微血管构筑的改变。方法采用KA癫痫模型(颈部皮下注射KA,10mg/kg),在造模后7天应用碱性磷酸酶法显示海马脑片片厚(90μm)的微血管,光镜观察,使用NIS-Element BR软件定量分析。结果海马内的微血管成层分布,构筑模式与神经元的构筑模式相一致;KA组的微血管数目明显多于对照组(P=0.003),血管平均长度明显高于对照组(P=0.000),血管平均直径无明显变化(P=0.121)。结论海马齿状回分子层微血管构筑在癫痫发病早期发生改变。 Objective The quantitative observation of the microvessels in dentate gyrus molecular layer of hippocampus was carried out to explore the change of the angioarchitecture of the hippocampus in the epilepsy rats.Methods The rats induced by kanic acid(KA)subcutaneous injection(10mg/kg)were used as the animal model for epilepsy,and the microvessels of hippocampus were stained by the alkaline phosphatase method at the 7th day after KA injection.The slices of the hippocampus with 90μm thick were observed by a light microscope and analyzed quantitatively by NIS-Element BR analysis system.Results The microvessels of the hippocampus were arranged in layers consistent with the pattern of the neurons of the hippocampus.The number of microvessels in dentate gryrus molecular layer was significantly increased in KA group than in control group(P=0.003),and the average length of microvessels in KA group was longer significantly than in control group(P=0.000).However the average diameter of the microvessels in two groups was similar(P=0.121).Conclusion The alteration of the microvascular network in molecular layer of the hippocampus happened in early stage of epilepsy.
