摘要
目的:研究LKB1、AMPK在人胃癌组织中的表达情况及其在胃癌发生中的作用机制。方法:收集胃癌组织、癌旁组织和癌旁正常组织各30例,采用实时荧光定量PCR法检测胃癌组织中LKB1 mRNA含量;用蛋白印迹法和免疫组化法检测胃癌组织中LKB1及其下游信号AMPK的表达状态。结果:胃癌组织中的LKB1 mRNA水平显著高于癌旁正常组织,而LKB1蛋白表达量和AMPK的172位苏氨酸(Thr)的磷酸化水平显著低于癌旁正常组织。结论:在胃癌组织中,肿瘤抑制因子LKB1的表达可能存在转录和(或)翻译后的调节,而LKB1蛋白激酶活性显著降低,提示存在LKB1/AMPK信号通路下调。
Objective To study the expressions of tumor suppressor gene LKB1 and its product in gastric cancer.Methods Real-time fluorescence quantitative PCR was used to detect LKB1 mRNA in cancerous tissue,paraneoplastic tissue and adjacent normal gastric tissue specimens obtained from 30 patients with gastric cancer;Western blot and immunohistochemistry(IHC) were used to detect the expressions of LKB1 protein and its downstream signal protein AMPK.Results The level of LKB1 mRNA in cancer tissue is significantly higher than that in adjacent normal gastric tissue specimens(P0.05),and the level of LKB1 protein and activity of AMPK were significantly lower than that in adjacent normal gastric(P0.05).Conclusions Transcription and post-translation modulation of LKB1 expression may exist in gastric cancer tissue,and the decrease of LKB1 protein kinase activity suggests the down-regulation of LKB1 /AMPK signal pathway.
