摘要
目的:建立用HPLC法测定清火片中大黄素和大黄酚的含量及含量均匀度的方法。方法:采用phenomenex HyperClone柱(C18,4.6×250mm,5μm),以流动相为甲醇-0.1%磷酸溶液(85∶15),流速为1.0ml.min-1,检测波长为254nm,进样量为20μl;柱温为室温。结果:大黄素和大黄酚分别在0.5～5.5μg.ml-1(r=0.9995)和5～50μg.ml-1(r=0.9992)范围内,进样浓度与峰面积呈良好线性关系,大黄素和大黄酚的平均回收率分别为96.8%和99.4%,RSD为0.36%和0.48%(n=5)。结论:该方法简便、准确,可作为清火片质量控制的方法。
Objective： To establish method for determining the content and uniformity of Emodin and Chrysophanol in Qinghuo Tablets by HPLC. Methods： The Phenomenex HyperClone （C18, 4. 6 × 250mm, 5μm） was used and mobile phase was methanol -0. 1% Phosphoric acid （85： 15） , and its temperature was used at room temperature, with the flow rate of 1. 0ml·min-1and the wavelength of detection of 254 nm, and the injection amount of 201xl. Results： The linear ranges of emodin and chrysophanol were at 0. 5 - 5.5μg·ml-1 （ r = 0. 9995 ） and 5 - 50μg·ml-1（ r = 0. 9992 ）, the average recoveries （ n = 5 ） were 96. 8% and 99.4% with RSD 0. 36% , 0. 48%. Conclusions： The method was simple, accurate, and can be used for the quality control of Qinghuo tablets.
