摘要
目的考察黄芪、丹参、三七醇提药渣中粗多糖提取工艺。方法采用正交试验,考察不同溶剂用量、提取时间、提取次数对多糖得率的影响,优选提取工艺;考察采用不同药液浓度、醇沉液浓度、沉淀时间所得多糖总量与纯度,优选沉淀工艺。结果优选出的多糖提取工艺为以10倍量水提取2次,每次1.5 h;优选出的沉淀工艺为药液浓度为1 g原药材/mL,醇沉浓度为80%,沉淀时间为4 h。结论优选出的工艺比较适合工业化生产的要求,具体的工艺技术参数将在中试研究中确定。
Objective To investigate the technological parameters of extracting crude polysaccharide from the mixture dregs of a decoction containing Radix astragali,Radix salviae miltiorrhizae and Radix notoginseng.Methods Orthogonal experiment was used to evaluate the solvent volume,extracting time as well as times that affect the yield of the crude polysaccharide.Also the alcohol precipitation technology was investigated by estimating the amounts and purity under the conditions of various specific gravity,concentration of alcohol and settling time.Results The optimized technological parameters were as fllows:the mixture was extracted twice by water as 10 times(w/w) for 1.5 h each time,then combined and filtered the water solutions,concentrated to equivalently 1 g(crude drugs)/mL,afterwards added the alcohol up to the concentration 80%,4 h later,the sediment was obtained.Conclusion The parameters were suitable for the actual production condition,and the confirmed parameters would be achieved by pilot study.
