摘要
目的了解低氧诱导因子1α(hypoxia-inducible factor-1alpha,HIF-1α)在急性高眼压大鼠视网膜中的表达,并探讨其在急性青光眼损伤中的作用。方法 80只Wistar大鼠随机分为对照组、穿刺组、加压后2h、12h、1d、3d、7d、14d组。对照组不做任何处理,穿刺组只进行前房穿刺不加压,加压各组前房灌注加压法制作急性高眼压模型。HE染色观察视网膜的形态学变化,免疫组织化学方法和RT-PCR方法观察HIF-1α蛋白和mRNA在视网膜中的表达情况。结果急性高眼压损伤后1d,光学显微镜下可见视网膜高度水肿,7d时水肿基本消失,14d时视网膜各层结构紊乱,明显萎缩变薄。HIF-1α蛋白和mRNA在加压后2h时表达增多(HIF-1α蛋白IOD值0.54±0.06,mRNA相对表达量1.5162±0.1199),12h达到高峰(HIF-1α蛋白IOD值1.73±0.10,mRNA相对表达量2.9041±0.1690),随时间推移表达逐渐减少,14d时降至最低(HIF-1α蛋白IOD值0.52±0.03,mRNA相对表达量1.2847±0.0434),但仍高于对照组(HIF-1α蛋白IOD值0.25±0.04,mRNA相对表达量1.0009±0.0480),差异均有统计学意义(均为P<0.05)。结论大鼠视网膜的HIF-1α在急性高眼压后表达增加,该因子在急性青光眼发病机制中具有重要作用。
Objective To investigate the expression of hypoxia-inducible factor-1 alpha(HIF-1α) in rat retina after acute intraocular hypertension,and discuss the effect of HIF-1α in retinal damage under acute hypertension.Methods Eighty Wistar rats were divided randomly into control group,punctured group,2 hours,12 hours,1 day,3 days,7 days and 14 days after ocular hypertension groups.Rats in control group were without any treatment,only anterior chamber puncture was taken in punctured group,and acute intraocular hypertension model in ocular hypertension group was created by anterior chamber perfusion.The histological change of retina was observed by HE staining.The expression of HIF-1α in retina was determined by RT-PCR and immunohistochemistry.Results Under optical microscope,the retina was highly edematous at 1 day after acute intraocular hypertension,the edema disappeared at 7 days and the retina was atrophic and thinning at 14 days.The expression of HIF-1α increased at 2 hours after acute intraocular hypertension(IOD:0.54±0.06,mRNA:1.516 2±0.119 9),reached to peak at 12 hours(IOD:1.73±0.10,mRNA:2.904 1±0.169 0)and reduced to the minimum at 14 days(IOD:0.52±0.03,mRNA:1.284 7±0.043 4),but still higher than those in control group(IOD:0.25±0.04,mRNA:1.000 9±0.048 0),there were statistical differences(all P〈0.05).Conclusion The expression of HIF-1α increases in rat retina after acute intraocular hypertension,and HIF-1α plays an important role in the pathogenesis of acute glaucoma.
