摘要
建立了鱼粉中组胺的柱前衍生、液相色谱检测方法。样品经高氯酸溶液提取、丹磺酰氯衍生化反应后经高效液相色谱—紫外检测器检测,外标法定量。方法的检出限为50 mg/kg,浓度在2.0~80.0μg/mL范围内,线性良好(R2=0.999 3),添加浓度在50、300、1 000 mg/kg时,回收率均大于70%,相对标准偏差(RSD)小于7%,结果表明该方法可适用于鱼粉中组胺的测定。
A pre-column derivatization and high performance liquid chromatography(HPLC) method was developed for determination of Histamine in fish meal.Samples were extracted in perchloric acid solution,followed by dansyl chloride derivatization reaction.Analytical identification and quantification were performed using HPLC equippmented with UV detector.The detection limit was 50 mg/kg.Good linearity between chromatographic peak area and Histamine concentration ranged from 2.0 to 80.0 μg/mL with the correlation coefficient of 0.999 3.Average recoveries of Histamne in fish meal spiked at the levels of 50,300,1 000 mg/kg were all more than 70%,with relative standard deviations of less than 7%.The results showed that thie method could be applied to the determination of histamine in fish meal.
