摘要
利用人胚胎及成熟组织做实验材料,通过Western杂交技术检测Rb基因的表达水平;藉以了解Rb基因与细胞生长、增殖及分化的关系。实验结果表明:(1)同一组织或器官随着月龄增加Rb蛋白的表达量逐渐增加;(2)不同的组织或器官Rb基因开始表达的时间顺序不同,Rb蛋白的表达量亦各不相同;(3)中年以后Rb基因的表达量明显降低。
In order to study the roles of the Rb gene in the control of growth and differentiation of cells we selected embryonic tissues as material to study Rb gene expression. The protein extraction was carried out by the procedure introduced by Prof. Wenhua Lee and was analysed by SDS-PAGE then immunoprecipitated with specific anti-Rb antibody. The results from western blot suggested that: (1) in most tissues the more mature the embryonic tissue the stronger the expression of Rb protein, such being most evident in retina, kidney, skin and lung; (2) there were differences in the expressive quantity from different tissues of the same foetus; in retina, kidney, skin, lung, heart and muscle, the Rb protein was obvious while in brain and liver it was weakly expressd; (3) the expressive quantity of Rb protein in a middle-aged man be markedly decreased even less than the 32-week foetus.
