摘要
目的 :探讨p16、Rb基因在骨肉瘤中的表达及意义。方法 :采用免疫组织化学方法 (SP法 ) ,检测了 2种基因在 4 3例骨肉瘤和 15例骨软骨瘤中的表达 ,并在 2种肿瘤之间及骨肉瘤不同分化组之间进行比较。结果 :抑癌基因p16在骨肉瘤中表达阳性率 (6 9 8% )显著高于骨软骨瘤组的阳性率 (2 0 % ) ,P <0 0 1;Rb基因在骨肉瘤中的表达阳性率 (5 5 8% )显著低于骨软骨瘤组的阳性率 (86 7% ) ,P <0 0 5。在骨肉瘤中 ,p16与Rb基因表达之间存在显著负相关 (r =- 0 6 0 5 ,P <0 0 1)。 2种基因表达率在骨肉瘤不同分化组间差异无显著性 (P >0 0 5 )。结论 :在骨肉瘤中 ,不同程度地存在p16、Rb基因的表达异常 ,其在骨肉瘤发生发展中起着重要作用 ,是骨肉瘤分子水平研究的重要标志物。
Objective To investigate the expression of p16 and Rb genes in osteosarcoma and their correlativity.Methods Expression of two kinds of genes in 43 osteosarcoma and 15 osteochondroma was detected by immunohistochemistry staining (SP).The comparison was made among two kinds of tumor and groups with differently differentiating extent in osteosarcoma.Results As a anti cancer gene,p16 gene positive rate was significantly higher in osteosarcoma (69.8%) than in osteschondroma (20%), P <0.01.The Rb gene positive rate was significantly lower in osteosarcoma (55.8%) than in osteochondroma (86.7%), P <0.05.In osteosarcoma,significant negative correlativity existed between the expression of p16 gene and Rb gene ( r =-0.605, P <0.01).No significant difference was observed among the positive rate of two kinds of gene expression in osteosarcoma groups with differently differentiating extent.Conclusions The abnormal expression of p16 Rb genes exists to different exent in osteosarcoma.Two kinds of genes play an important role in the occurrence and development.And they may be an important marker for molecular research of osteosarcoma.
