艾炷灸对D-半乳糖衰老小鼠学习记忆能力和大脑皮质、海马区c-fos蛋白表达的影响 被引量:10

摘要 目的观察艾炷灸对D-半乳糖衰老小鼠学习记忆能力的影响与学习记忆相关脑区c-fos蛋白表达的关系。方法健康3月龄昆明雄性小鼠体质量(30±2)g,60只,水迷宫检测后按随机区组设计分为:生理盐水组,造模组,艾灸1、2组,尼莫地平组等5组,每组各12只。造模组,艾灸1,2组,尼莫地平组连续42 d颈背部皮下注射D-半乳糖造成小鼠衰老模型;生理盐水组连续42 d颈背部皮下注射等量生理盐水,12 d后艾灸1、2组,分别选取小鼠"足三里、悬钟","关元、百会"行艾柱灸治疗,尼莫地平组给予尼莫地平灌胃,均为隔日1次,共15次。采用水迷宫检测学习记忆、免疫组化方法观察各组小鼠大脑皮层、海马CA3区c-fos蛋白的表达。结果模型组潜伏期较生理盐水组、艾灸1、2组、尼莫地平组延长(P<0.01或P<0.05),模型组原象比、垮台次数较其他各组缩短、降低(P<0.01或P<0.05),模型组小鼠大脑皮质、海马CA3区c-fos蛋白表达的神经元密度较生理盐水组、艾炷灸组、尼莫地平灌胃治疗组降低(P<0.01)。结论艾炷灸足三里、悬钟,关元、百会穴位处方可能通过提高D-半乳糖衰老小鼠脑组织大脑皮层、海马CA3区c-fos蛋白的表达改善学习记忆能力。 Objective To observe the influence of moxa cone moxibustion on learning and memory function and expression of c-fos protein in the related cerebral tissue in senile mice induced by D-galactose injection.Methods 60 male mice(3 months old) were randomly and equally divided into the normal sodium control,model,moxa cone moxibustion 1(acupoint ST36 and GB39),moxa cone moxibustion 2(acupoint DU20 and RN4),Nimodipine medicated groups.The treatment and model groups were given with D-galactose injection for 42 days,while injected the same dosage of normal sodium in the normal sodium control.From the 13th day on the first injection,the three treatment groups were treated every other day for one month.The spatial learning and memory was measured with the morris water maze(MWM) computer system and the expression of c-fos protein on the cerebral cortex and hippocampus(CA3) of senile mice was detected with immunohistochemical method.Results The significant difference was observed between the modele group and the other four groups on spatial learning and memory,the escape lantency in model group was prolonged,the spanning(original) platform frequency,the percentage of(original) platform quadrant were decreased(P0.01;P0.05).The expression of c-fos protein in cerebral cortex and hippocampus(CA3)in model group was decreased compared with other groups(P0.01).Conclusion The moxa cone moxibustion prescription on ST36 and GB39,DU20 and RN4 can improve learning and memory function of senile mice induced by D-galactose,and the mechanim may be related to the increasing expression of c-fos protein in cerebral cortex and hippocampus(CA3).
