摘要
目的:用屋尘螨提取液(House DustMite Extract,HDM)构建哮喘小鼠气道重塑模型,检测肺组织中α平滑肌肌动蛋白(Smooth Muscle Actin-α,α-SMA)的表达。方法:BALB/c小鼠随机分为空白组、HDM低、高剂量三组,每组再分为3、6、9周3个亚组。用不同浓度的HDM皮下/腹腔致敏,滴鼻激发。用医学图像软件分析气道重塑的病理改变,分别用qRT-PCR和IH检测肺组织中α-SMAmRNA和蛋白的表达。结果:与空白组对比,HDM高、低剂量组肺组织病理炎症计分增加,气道内外径比值降低,α-SMA蛋白及其mRNA表达增加,随激发时间延长改变越来越显著;高剂量组较低剂量组更显著。结论:HDM诱导BALB/c小鼠支气管哮喘气道重塑中,α-SMA随气道重塑的加剧而表达增加。
Objective:To build an airway remodeling model of bronchial asthma in mouse with house dust mite extracts,to investigate the expressions of α-SMA in lungs,and clarificate the relationship between them.Methods:BALB/c mice were randomly divided into control group,HDM low-dose group and HDM high-dose group.Each group was randomly divided into three subgroups:three-week,six-week and nine-week group.The mice were challenged by intranasal drip after sensitized with subcutaneous and peritoneal injection of HDM with different doses.Technique of computer-assisted image analysis was used to detect the changes of the airway remodelling pathomorphology.The expressions of α-SMA in lungs were detected by immunohistochemistry(IH) and Real-time quantitative reverse transcription polymerase chain reaction(qRT-PCR).Results:Compared with control groups,in both high-dose HDM and low-dose HDM groups,the histopathological scores increased;the ratio of inner and outer diameter(D1/D2) was notably decreased;The expression of α-SMA protein and mRNA increased significantly;and all had changed severely with the prolongation of exposured time.Compared with high-dose HDM groups,low-dose groups changed lighter.Conclusion:The expression of α-SMA mRNA and protein in asthmatic mice airway remodeling induced by HDM was increased with the airway remodeling.
