摘要
目的探讨重金属铬离子对孕ICR小鼠胚胎以及仔鼠的发育影响,为行人工全髋关节表面置换术后重金属铬离子对妊娠机体可能产生的影响提供实验依据。方法将56只雌性受孕ICR小鼠随机分成高、中、低剂量组及对照组4组,每组14只。实验组以不同剂量重铬酸钾灌胃染毒,对照组予双蒸水灌胃。36只于孕19 d处死,观测着床数、黄体数、胎窝总重、胎盘总重、活胎数、死胎数。检测另20只母鼠娩出的仔鼠活产数、雌雄比、体重、身长、尾长。42 d处死仔鼠,测定其血清铬和肝脏铬含量,取肝脏组织作病理学观察。结果随染毒剂量的增加,中、高剂量组着床总数与对照组相比呈减少趋势,高剂量组胚胎存活率低于对照组,胚胎死亡率高于对照组。高剂量组仔鼠血清、肝脏铬含量高于中、低剂量组,差异有统计学意义(P<0.01),高剂量组仔鼠肝脏细胞内出现大颗粒脂滴、染色体浓聚、线粒体减少的变化。结论本实验提示低剂量铬对胎鼠、仔鼠发育无影响,高剂量金属铬离子对孕鼠胚胎及仔鼠的发育产生较显著的影响,在子代体内仍存在毒性效应。
Objective To investigate the effects of chromium ions on embryos of pregnant ICR mouse and the development of offspring,and to provide experimental basis for the possible impact on pregnancy of chromium ions after hip surface replacement.Methods 56 pregnant female ICR mice were divided into high,medium and low-dose groups as well as control group,14 mice in each group.The experimental groups were fed with different doses of potassium dichromate and treated,while the control group were fed with distilled water.36 mice were sacrificed on the 19th day of pregnant.Implantation,luteal number,total litter birth weight,placental weight,number of living births,and stillbirths number were measured.As for the other 20 female Wistar mice,living birth number of offspring,sex ratio,body weight,body length,and tail length were observed and recorded.On the 42nd day,pups were sacrificed;the content of chromium in liver and serum was determined,and the liver tissue was taken for pathological examination.Results With the dose increased,the total number of implantation in the high dose group tended to decrease,compared with the control group.The high dose group had a lower embryo survival rate and higher embryo mortality than control group.Chromium content of serum and liver was higher in the high dose group than in the low dose group,and the difference was significant(P0.01).Large particles appeared in the lipid droplets of liver cells in high dose group offspring,as well as chromosome concentration,and mitochondrial number reducing.Conclusion This study suggested low-dose chromium has no effect on offspring growth,high-dose chromium ions have more significant impact on pregnant mice and the development of offspring,and as for the offspring,the toxic effects still exist in vivo.
