摘要
建立青蒿素抗疟相关化合物东莨菪内酯、猫眼草酚及猫眼草黄素的同时提取及其含量测定方法。采用乙醇回流提取3种化合物,HPLC-UV测定其含量,结果表明:乙醇回流提取3个目标化合物的回收率均在97.83%-99.48%之间,相对标准偏差CRSD〈5.00%);3种化合物在各自的检测限内线性关系良好(r≥0.999),该方法的稳定性(RSD〈5.00%)、重现性(RSD〈5.00%)及精密度(RSD〈5.00%)良好。该方法简便、快速、线性范围宽,可用于青蒿药材及提取物中3种化合物的质量控制;特别适宜不同生长阶段青蒿样品中东莨菪内酯、猫眼草酚及猫眼草黄素含量的跟踪监测。
A method can simultaneously determine the scopoletin, chrysosplenol-D and chrysosplenetin in leaves of Artemisia aunua was established. Three compounds were extracted with ethanol reflux, and analyzed by HPLC-UV system. The results showed that recovery rate of target compounds were 90%-105% and relative standard division was below 5.00%. Three compounds had good linear relationship (r ≥0.999) in their linear range under conditions of HPLC-UV. The method had good stability (RSD〈5.00%), reproducibility (RSD〈5.00%) and precision (RSD〈5.00%). Therefore, the method was simple, rapid, accurate and reproducible, which could be applied in the quality control of three compounds of crude drugs or extracts of A. atmua. In addition, it could be used to detect the contents of scopoletin, chrysosplenol-D and chrysosplenetin in leaf samples of A. annua during different growth stages.
