荧光增白剂对鳃金龟围食膜的影响及其对B.t.的增效作用

Effect of Fluorescent Brightener on Peritrophic Membrance of Melolonthid Larvae and Its Enhancing Effect on B.t.

摘要 [目的]B.t.防治鳃金龟幼虫存在杀虫效果慢、幼虫死亡率低的缺点,试验目的是提高B.t.对鳃金龟幼虫的杀虫活性。[方法]应用荧光增白剂FB28处理华北大黑鳃金龟和暗黑鳃金龟围食膜,并通过FB28与B.t.对暗黑鳃金龟进行生物测定。[结果]荧光增白剂FB28能解离华北大黑鳃金龟和暗黑鳃金龟围食膜,分别至少有11条和8条蛋白多肽被解离,且随处理时间的增长,蛋白被解离的量增大,围食膜被破坏程度增大。FB28与B.t.一起使用,对其具有明显的增效作用。[结论]围食膜蛋白主要是几丁质结合蛋白,FB28可使几丁质结合蛋白从围食膜上解离下来,FB28对B.t.杀虫具有增效作用,为应用荧光增白剂作为增效因子防治鳃金龟幼虫提供了理论依据。 [Aims] The aim was to enhance the activity of B.t. to control melolonthid larvae because of its defect of slow effect. [Mothods] The disruptive effect of fluorescent brightener 28 (FB28) on the peritrophic membrane (PM) of melolonthid larvae and its enhancing effect on B.t. virulence were studied through SDS-PAGE, structural observation and bioassay. [Rosults] FB28 could destroy the PM ofHolotrichia oblita and H. parallela and at least eleven and eight protein bands were dissociated from the two PMs respectively. With the growth of time, an amount of proteins were released and the PMs were then greatly damaged. The bioassay with H. parallela showed that FB28 had a synergistic effect on Rt.. [Gonclusions] FB28 could dissociate the proteins from PMs, most of which were chitin-binding proteins and could enhance the activity of B.t. significantly, which was theoretically helpful for further study on the application of fluorescent brightener as enhancing factor to control melolonthid larvae.
