摘要
应用光学显微镜和生化技术,研究了华北大黑鳃金龟(Holotrichia oblita)围食膜的基本结构和表面形态,初步分析了围食膜蛋白质的种类,蛋白质和糖的含量,测定了棉铃虫颗粒体病毒增效蛋白对围食膜结构的影响。结果表明,华北大黑鳃金龟围食膜表面平滑致密,富有弹性,但与多数昆虫围食膜相比韧性较差,主要由蛋白质、糖和几丁质组成,其中蛋白质的含量约为36.25%,糖的含量约为8.91%,其他为几丁质等;经SDS—PAGE测定,围食膜的蛋白质种类较多,有28条多肽比较清晰,分子量多在17.6—320.3kDa。棉铃虫颗粒体病毒增效蛋白不能降解华北大黑鳃金龟围食膜中的蛋白质。
As an initial step to study the structure of PM from Holotrichia oblita, the basic structure of the PM was analyzed by microscopy and the PM morphological aspects were observed by biochemical technology. The results showed that major components of the PM were proteins accounting for 36.25 %, and carbohydrates accounting for 8.91% by weight besides chintin. PM surface appeared to be smooth, close and highly elastic. Apparently 28 polypetides were separated by SDS-PAGE analysis with molecular weight from 17.6 to 320.3 kDa. Heliothis armigera granulosis virus enhancin could not degraded any proteins by assay in vitro.
