摘要
探讨不同的稀释介质与稀释方式对化学发光免疫法测定血清雌二醇(Estradiol,E2)浓度的影响。评价SIEMENSCentaur化学发光分析仪的E2测定精密度与线性范围。然后选择线性范围内的血清样品,等分为4份,1份使用仪器自动稀释的方式和配套稀释液D9完成测定,另外3份分别使用配套稀释液D9、混合低值血清和生理盐水作为稀释介质进行手工稀释5倍后上机测定。计算稀释测定结果与原始值的差异、相关系数,拟合回归方程。结果显示:用配套稀释液D9、混合低值血清和生理盐水进行稀释后测定E2的结果与原始值相比,差异无统计学意义(均有P>0.05);使用仪器自动稀释方式和配套稀释液得到的结果与原始值偏差最小。结论:采用仪器自动稀释方式和配套稀释液测定高值E2浓度为最适合的稀释方案。
To explore the influence of dilution medium and method on detection of serum estradiol concentration using chemiluminescence immunoassay,the precision and linearity of the analysis system was evaluated and verified before the study.Twenty-seven serum samples which the estradiol concentration was within the linear range were selected for the dilution experiment.Each sample was divided into four parts,one was measured for estradiol with automatic dilution using SIEMENS Centaur with assorted dilution medium D9,while the other three parts were diluted manually by assorted dilution medium D9,mixed low-level estradiol serum and normal saline respectively,and then measured by SIEMENS Centaur.The diluent fold was five for all parts.The results of diluted estradiol were collected and analyzed,and they were compared with the original values.Paired t-test was applied to observe the difference of dilution group and original group.The relative bias,correlation coefficient and linear regression were analyzed as well.The results showed that the diluted estradiol level was correlated well with the original value in all of the four groups.No significant difference was found in each dilution group and original group(P0.05).However,the lowest bias was obtained in the group of automatic dilution and assorted dilution medium.The best and most appropriate measurement of high level estradiol was using instrument automatic dilution mode and with the assorted dilution medium.
