摘要 目的检测survivin和p53蛋白在结直肠肿瘤中的表达,探讨其相互关系。方法收集结肠镜活检的结直肠腺瘤及结直肠腺瘤并发结直肠癌组织蜡块各30例,另取30例正常结直肠黏膜组织作对照,采用链霉菌抗生物素蛋白-过氧化酶连接免疫组织化学方法检测survivin和p53蛋白表达情况。结果 survivin、p53蛋白在结直肠腺瘤癌变过程中的表达随着腺瘤的癌变而增高,在结直肠腺瘤、结直肠腺瘤并发结直肠癌中的腺瘤及结直肠癌的表达率分别为26.7%、53.3%、66.7%和23.3%、56.7%、73.3%。从结直肠腺瘤到并发结直肠癌的腺瘤,survivin、p53蛋白的表达显著增高(P<0.05)。结论 survivin的异常表达与结直肠腺瘤癌变密切相关,在结直肠癌的发生中可能与p53起一定的协同作用。 Objective The aim of this paper is to investigate the expression of survivin and its correlation with expression of p53 protein in colorectal tumor tissues by immunohistochemistry, and to preliminarily evaluate the clinical significance of surviving and p53 in the carcinogenesis and progression of colorectal cancer. Methods Specimens for this study were composed of 30 colorectal adenoma tissues, 30 cancerous adenoma tissues, all of which were resected endoscopically, 30 normal colorectal mucosal tissues were used as control. The immunohistochemical staining of survivin and p53 was carried out with the standard streptavidin-peroxidase(SP) technique. Results During colorectal tumorigenesis, nuclear accumulation of survivin and p53 protein was increased gradually in the malignant transformation within adenomas. The positive rates for survivin and p53 protein in colorectal adenomas, colorectal adenomas complicated by carcinomas and colorectal carcinomas were 26.7%、53.3%、66.7% and 23.3%、56.7%、73.3%, respectively. Colorectal adenomas comparing with colorectal adenomas complicated by carcinomas, the immunoreactivitie of p53 protein was increased significantly (P〈0.05). Conclusions The aberrant expression of survivin in colorectal adenoma was closely correlated with malignant transformation. It might cause synergistic effect with the mutation of p53 during colorectal tunorigenesis.
