电穿孔过程中细胞膜电导率变化条件下的跨膜电压研究 被引量:3

Transmembrane Voltage Studies with Consideration of the Membrane Conductivity Variations in the Process of Electroporation

摘要 基于经典球形单细胞五层介电模型,并考虑电穿孔过程中细胞膜电导率的变化,对微秒脉冲电场作用下的细胞内外膜跨膜电压进行了仿真分析,以及脉冲电场诱导人宫颈癌细胞系Hela凋亡的实验研究。仿真分析发现,随着细胞穿孔程度的加剧,细胞膜电导率增加,内、外膜跨膜电压的比值不断增加,表明外加脉冲电场对内膜的影响不断增强,其作用靶点逐渐透过细胞膜进入胞内细胞器。同时,实验结果也表明随着脉冲电场场强不断增加,微秒脉冲电场所诱导的Hela细胞的凋亡率随之增加,从而间接证实了仿真分析的结果。现有研究普遍认为微秒脉冲电场的作用靶点为细胞外膜,本研究关于微秒脉冲电场对细胞内膜作用的研究结果是对已有研究的重要补充和发展。 Based on the classical 5-layer singular cell model,this paper simulated both the outer and the inner transmembrane voltages induced by microsecond-pulsed electric field applied by taking into consideration the membrane conductivity variations in the process of electroporation.It is found that with the extent of cell electroporation increasing,the ratio of the peak value of the inner transmembrane voltage to the outer membrane increases due to the variations of membrane conductivity.The increased ratio implies an increasing effect imposed on the inner membrane by the field and a target-spot transfer of the field from the outer membrane to the inner.Finally,an apoptosis experiment on Hela cells was conducted and a similar conclusion to the simulation analysis above was deduced.It is generally accepted that the target-spot of the microsecond pulsed electric filed is the outer membrane and thus the conclusion of this paper is an important supplement and development of current studies.
