摘要
研究脂多糖(LPS)体内诱导小鼠胸腺细胞凋亡,发现胸腺细胞DNA凝胶电泳可出现凋亡特征的梯状带,并且胸腺细胞的凋亡与LPS剂量呈依赖关系;苏木精、伊红染色后镜下可见胸腺皮质部分出现大量凋亡细胞,但难以计数;终末脱氧核苷酸转移酶介导的脱氧尿嘧啶三磷酸腺苷原位缺口末端标记法可计数胸腺皮质凋亡的细胞,该法观察LPS体内诱导胸腺细胞凋亡的动力学特征表明,在注射LPS后12~18h之间胸腺细胞凋亡的百分率迅速增加.
Apoptosis of murine thymocytes induced by lipopolysaccharide (LPS)in vivo was investigated. Induction of DNA fragmentation in thymus by administration of LPS to mice was dependent on the dose of LPS. The quantitative assay for apoptosis in thymus tissue stained with H&E was difficult. Apoptosis of murine thymocytes in the cortex in vivo was countable by using terminal deoxynucleotidyl tranferase-mediated dUTP nick end labeling (TUNEL). The apoptosis was mostly occurred from 12 h to 18 h after injection of LPS.
