姬松茸多糖对铅中毒大鼠胸腺Bax和Caspase-3表达的影响 被引量:4

EFFECTS OF POLYSACCHARIDES FROM AGARICUS BLAZEI MURILL ON THE EXPRESSION OF BAX AND CASPASE-3 IN THYMUS OF LEAD-POISONING RATS

摘要 目的了解姬松茸多糖对铅中毒大鼠胸腺Bax和caspase-3基因表达的影响。方法选择健康45 d龄SD大鼠48只,随机分为6组,雌雄各半,分别为正常对照组、多糖对照组,Pb中毒模型组和Pb中毒+低、中、高剂量多糖共6组。铅中毒模型组和低、中、高剂量组给予0.2%醋酸铅溶液,其余组给予蒸馏水,自由饮用。多糖对照组、低、中及高剂量组分别按每天每只100、50、100、200 mg/kg体重灌服姬松茸多糖液,各组灌服液体积相等,均为1ml。其它组每天每只灌胃1ml的生理盐水。饲养60d后,取大鼠胸腺,通过荧光定量PCR测定Bax和caspase-3基因表达量。结果 (1)与正常对照组相比,胸腺Bax和caspase-3的表达量,铅中毒模型组分别升高了17%和14%差异,具有统计学意义(P<0.05);多糖对照组分别降低18%和31%,差异有统计学意义(P<0.01或P<0.05)。(2)与铅中毒模型组比,三个剂量组中多糖剂量越高,表达量越低,其中高剂量组Bax和caspase-3的表达量分别降低了44%和60%,差异具有统计学意义(P<0.01)。结论姬松茸多糖对铅致大鼠凋亡基因Bax和caspase-3表达增强具有抑制作用。[营养学报,2012,34(6):582-585,590] Objective To observe the effects of polysaccharides from Agaricus blazei Murill on the expression of Bax and caspase-3 in the thymus of lead-poisoning rats.Method Forty-eight SD rats aged 45 d(female:male=1:1) were randomly divided into normal control,lead-poisoning model,polysaccharides control,lead+low-dose polysaccharides,lead+medium-dose polysaccharides and lead+high-dose polysaccharides groups.Lead-poisoning group and three Lead+polysaccharidc groups were given 0.2% lead acetate in drinking water,and the others with distilled water.Polysaccharides control group,low,medium and high dose polysaccharides groups were treated with 100,50,100 and 200 mg/kg bw polysaccharides from Agaricus blazei murill every day respectively and the others were administered the same capacity of nornral saline(0.9%).After 60 d,the thymus was collected.The expression of Bax and caspase-3 was determined with fluorescent quantitative real-time polymerase chain reaction(FQRT-PCR).Results(1)Compared with the normal control group,the expression of Bax and caspase-3 in lead-poisoning model group was increased respectively by 17% and 14%(P 0.05),and decreased respectively in polysaccharides group by18% and 31%(P0.01 and P0.05).(2) Compared with the lead-poisoning model group,the expression of Bax and Caspase-3 was decreased significantly in high dose polysaccharides group by 44% and 60%(P0.01).Conclusion Polysaccharides from Agaricus blazei Murill can inhibit mRNA expressions of Bax and caspase-3 in lead-poisoning rats.[ACTA NUTRIMENTA SINICA,2012,34(6):582-585,590]
