摘要
目的:研究槲皮素-7-硫酸酯钠盐(SQMS)对血栓形成、全血血小板聚集和血小板释放一氧化氮(NO)的影响。方法:用血栓生成仪观察大鼠颈动脉血栓形成情况;小鼠尾静脉注射胶原蛋白-肾上腺素混合诱导剂造成瘫痪模型,观察15min后小鼠瘫痪或死亡数;全血与药物孵育5min,然后用凝血酶或ADP作用1min,用全血血小板聚集仪观察全血血小板聚集情况;血小板悬液与药物孵育5min,然后用凝血酶作用1min,用NIKON TE2000-E荧光倒置显微镜观测血小板聚集情况;用硝酸还原酶法观察血小板释放NO情况。结果:SQMS 10mg/kg和20mg/kg抑制大鼠颈总动脉血栓形成,其抑制率分别为72%和92%;SQMS 20mg/kg和40mg/kg对混合诱导剂造成小鼠瘫痪或死亡有抑制作用,其抑制率分别为70%和80%;SQMS 160、320和640μmol/L抑制凝血酶和ADP诱导的全血血小板聚集,其抑制率分别为24%、76%5、7%和26%5、6%、38%;荧光倒置显微镜观测到血小板聚集的抑制作用随浓度的加大而增强;SQMS 80、160、320和640μmol/L对凝血酶诱导血小板释放NO有加强作用,NO的增加率分别是55%、118%、219%和394%。结论:SQMS对血栓形成有抑制作用,其作用机理与其抑制全血血小板聚集和增加血小板释放NO有关。
Objective:To study the effect of sodium quercetin-7-sulfate(SQMS) on thrombosis,platelet aggregation in whole blood and release of nitric oxide(NO) in platelets.Methods:The thrombosis in the carotid artery of rat was observed by a thrombosis inducer.Thrombosis of mice was induced by tail intravenous injection of a mixture of collagen and adrenaline and the number of paralysis or dead after 15 min was also calculated.Whole blood was incubated with SQMS for 5 min,and then was treated with thrombin or ADP for 1 min.Platelet aggregation was analysed by whole blood aggergometers.Suspensions of platelets were incubated with SQMS for 5 min,and then were treated with thrombin for 1 min.Platelet aggregation was observed by inverted microscope(NIKON TE2000-E).Release of NO in platelets was measured by nitrate reductase-dependent assay.Results:SQMS 10mg/kg and 20mg/kg inhibited thrombosis in the carotid artery of rat with inhibition rate 72% and 92%,respectively.SQMS 20mg/kg and 40mg/kg could inhibit the number of paralysis or dead induced by the mixture of collagen and adrenaline with inhibition rate 70% and 80%,respectively.SQMS 160、320 and 640μmol/L inhibited platelet aggregation in whole blood induced by thrombin and ADP with inhibition rate 24%、76% 、 57% and 26%、56%、38%,respectively.Enhanced inhibition on platelet aggregation with incremental concentration of SQMS was observed by inverted microscope.SQMS 80、160、320 and 640μmol/L had enhanced release of NO in platelets induced by thrombin with NO increase rate 55%、118%、219% and 394%,respectively.Conclusion:SQMS showed inhibition on thrombosis and its molecular mechanism was due to its inhibition on platelet aggregation in whole blood and release of NO in platelets.
