大鼠锥体束损伤对脊髓运动神经元神经生长因子表达的影响 被引量:3

Influence of the pyramidal tract injury on NGF and its mRNA in motor neurons of adult rats lumbar spinal cord

摘要 目的 探讨成年大鼠锥体束损伤后 ,腰段脊髓运动神经元 NGF m RNA及 NGF的含量变化。 方法 选择性毁损左侧锥体束 ,用原位杂交的方法标记 NGF m RNA,以 PAP法进行 NGF的免疫组化染色。并采用计算机辅助图像分析系统对切片进行分析、比较。 结果 正常成年大鼠腰段脊髓前角运动神经元中几乎不含 NGF m RNA及 NGF,左侧锥体束损伤后 1周 ,双侧运动神经元内 NGFm RNA及 NGF的含量均有明显增高 ,且右侧高于左侧。 结论 成年大鼠左侧锥体束损伤后 ,双侧腰段脊髓运动神经元中 NGF m RNA及其蛋白表达产物均较正常对照组有明显增高。 Objective To measure the changes in the contents of NGF mRNA and its protein products in lumbar motor neurons after the adult rat pyramidal tract was damaged, Methods The lumbar spinal cords of the rats were dissected periodically after the operation of the left pyramidal tract damage The NGF mRNA was detected with the in situ hybridization,and NGF was stained with immuno histo chemical method The slices were analyzed with the aid of computerized image analysis system Results The contents of both NGF mRNA and NGF in the lumbar motor neurons of normal adult rats were extremely low After damage of the left pyramidal tracts,the levels of NGF mRNA and NGF increased significantly within one week Then the NGF expression tended to decrease until at least four weeks after operation And the contents of both on the right side were higher than those on the left side Conclusion When the left pyramidal tract of an adult rat is damaged,the contents of NGF and its mRNA in lumbar motor neurons rise And those on the right side increase more significantly
