摘要
目的观察施万细胞长期移植入中枢神经系统后是否存活。方法取大鼠施万细胞体外培养,一部分经5'溴脱氧尿嘧啶(BrdU)标记后移植至电针损伤的大鼠中脑网状结构,另一部分经Hoechst33342标记后置于PLGA支架内再移植至大鼠全横断脊髓损伤处,分别于移植后的8个月和11个月处死动物,行BrdU免疫组织化学染色及荧光显微镜观察。结果施万细胞移植入脑内8个月后仍可见较多BrdU阳性细胞,褐色椭圆形,并向大脑皮层迁移;移植于脊髓11个月后荧光显微镜下可见许多Hoechst33342阳性细胞,发蓝色荧光,形态不规则,在PLGA支架内及其头、尾端脊髓实质内都可见到。结论施万细胞移植入中枢神经系统后可长期存活,并向远端迁移。
Objective To investigate Schwann cells whether survival and migration after transplanted to central nervous system for a long-term. Methods The Schwann cells of rat were expended in vitro, the part of them were labeled with S'-bromodeoxyuridine (Br- dU) and transplanted to rat's middle brain injured by electric needle stimulus, the others were labeled with Hoechst 33342, seeded to PLGA scaffold, and transplanted to rat's transected spinal cord. 8 and 11 months later, rat brain and spinal cord were taken out re- spectively, examined by BrdU immunohistochemistry and fluorescence microscope. Results BrdU positive cells could be seen after 8 months and migrated toward cerebral cortex. Hoechst 33342 positive cells could be identified in scaffold and transected spinal cord after 11 months under fluorescence microscope. Conclusion Grafted Schwann cells can survive in central nervous system for a long- term and migrate toward distance.
