摘要
目的:探索化瘀祛痰复方矢志方对尿酸代谢的影响及机制。方法:以酵母粉每日15 g/kg灌胃及氧嗪酸钾每日600 mg/kg腹腔注射2周诱导SD大鼠高尿酸血症模型。设正常组、模型组、中药(矢志方)组及苯溴马隆组,给予相应干预。2周后,检测各组大鼠血清尿酸、肌酐、尿素氮、血清黄嘌呤氧化酶(XOD)活性、尿尿酸、尿β2-MG及观察肾组织病理变化。结果:与空白组比较,模型组血尿酸、24 h尿量、尿尿酸水平、24 h尿酸总排泄量和XOD活性明显升高,差异有统计学意义(P<0.01),而血尿素氮、血肌酐、尿β2-MG差异无统计学意义(P>0.05),说明该高尿酸血症模型成功建立。给药后,与模型组比较,中药组及苯溴马隆组血尿酸显著降低、24 h尿尿酸总排泄量显著增加、血清XOD活性显著降低,差异均有统计学意义(P<0.01);中药组和苯溴马隆组各指标差异均无统计学意义(P>0.05);肾组织病理显示两治疗组病变程度较模型组均减轻。结论:矢志方可以有效降低高尿酸血症模型大鼠血尿酸的水平并可促进尿尿酸的排泄,可降低血清XOD活性,可能为其降低血尿酸的机制之一。
Objective: To investigate the effects of "Shizhi Formula" of resolving stasis and dispelling phlegm on uric acid metabolism and its mechanism.Methods: The hyperuricemia model of Sprague Dawley(SD) rat was established by intraperitoneal injection of potassium oxonate(600 mg·kg-1·d-1) and intragastric administration with yeast(15 g·kg-1·d-1) for two weeks.All the rats were divided into normal group,model group,"Shizhi Formula" group and benzbromarone group with corresponding intervention for each group.Two weeks later,the levels of serum uric acid,creatinine,urea nitrogen,xanthine oxidase(XOD) activity,urine uric acid,and urine β2-MG were detected,and the pathological changes of renal tissue were observed.Results: Compared with the normal group,the levels of serum uric acid,24-hour urine volume,urine uric acid and the total excretion of 24-hour uric acid and XOD activity in model group significantly increased(P0.01),but there was no significant difference in serum urea nitrogen,serum creatinine and urine β2-MG between the two groups(P0.05).It was proved that the hyperuricemia model had been successfully established.After administration,compared with the model group,the levels of serum uric acid and XOD activity in "Shizhi Formula" group and benzbromarone group decreased significantly(P0.01),and the total excretion of 24-hour urine uric acid significantly increased(P0.01);while there was no statistical difference between "Shizhi Formula" group and benzbromarone group in all above indexes(P0.05);the pathological changes of renal tissue in "Shizhi Formula" group and benzbromarone group were milder than model group.Conclusion: "Shizhi Formula" can reduce the level of serum uric acid and promote the excretion of urine uric acid in hyperuricemia rats;meanwhile it can reduce serum XOD activity,which may be one of the mechanisms of reducing uric acid.
