摘要
热激蛋白是一类结构非常保守的分子伴侣类型的应激蛋白,在调控动、植物代谢和信号传导等方面发挥重要作用。本研究利用同源克隆法,筛选非亲和条锈菌小种CYR32侵染诱导的cDNA文库,分离获得1个HSP70家族基因的全长cDNA序列TaHSP70。序列分析发现,该cDNA片段包含1个2 412 bp开放阅读框,推测其编码包含691个氨基酸残基的蛋白质,属于质体间质亚族蛋白。半定量RT-PCR方法分析发现,在小麦幼苗受到低温、干旱和植物激素ABA胁迫处理后,TaHSP70表达受到抑制。利用条锈菌非亲和小种CYR32和白粉菌亲和小种混合菌系接种后以及高温胁迫条件下,TaHSP70表达量增加。说明小麦HSP70家族基因TaHSP70可能参与调控小麦抗逆和防卫反应过程。
Heat shock proteins are a kind of molecular chaperones,of which structure are highly conserved,and play essential roles in regulating various metabolic processes and molecular signal transductions in animals and plants.Through homologous PCR cloning method,a novel full-length cDNA encoding a HSP70 protein was obtained from a cDNA library of near-isogenic line Taichung 29*6/Yr5 inoculated with an incompatible race CYR32 of Puccinian striiformis,and was named as TaHSP70.Sequence analyses indicated that TaHSP70 contained an open reading frame of 2 412 bp in length,encoding 691 amino acids.Clustering analysis showed that TaHSP70 belonged to the plastid stroma group of HSP70 protein family.We used semi-quantitative RT-PCR to analyze its expression patterns under biotic or abiotic stress conditions.It was found that TaHSP70 was suppressed by exogenous application of ABA,treatments of dehydration and cold,but it was induced by infection of incompatible strain CYR32 and compatible mixed races of powdery mildew and heat.Based on the expression pattern analysis,we proposed that TaHSP70 may participate in the regulatory processes of wheat defense and stress responses.
