摘要
热休克蛋白70(HSP70)广泛分布于生物体中,在蛋白转运、耐热性、细胞保护等方面起着重要作用。本研究以黑腹果蝇的一个HSP70氨基酸序列与家蚕基因组预测的基因库进行tBlastn检索,共获得了多个可能的家蚕HSP70基因,我们对编号为BGIBMGA002381进行了电子克隆。BGIBMGA002381基因的编码区序列与家蚕ESTs数据库进行Blastn比对检索,共获得了20条ESTs证据,经SeqMan拼接获得家蚕HSP70的cDNA序列长2057 bp,其编码区长1950 bp,共编码649个氨基酸。该HSP70蛋白质分子质量为71.18 KD,等电点为5.33。结果表明该HSP70基因在mRNA水平有表达,可能为家蚕基因组中功能性的HSP70,这为深入研究该基因的功能奠定了基础。
Heat shock protein 70(HSP) is widely distributed in organisms and plays important roles in protein transport,heat resistance and cell protection.In this study,an HSP70 amino acid sequence of Drosophila melanogaster was used to do Blastp search in the silkworm genome.Several putative HSP70 genes were found.We chose one of the gene(accession number: BGIBMGA007950) for electronic cloning.The putative coding sequence of BGIBMGA002381 was used to do Blastn search in the silkworm expressed sequence tags(ESTs) database.A total of 20 ESTs were obtained and assembled by SeqMan.The cDNA sequence of BGIBMGA002381 was 2057 bp in length.We predicted that the coding sequence of BGIBMGA002381 was 1950 bp in length,coding 649 amino acids.Its molecular weight and isoelectric point were 71.18 kD and 5.12,respectively.The results suggested that BGIBMGA002381 might be an active gene in the silkworm genome.
