不同种属、不同剂量甲状腺球蛋白诱导CBA/J小鼠实验性自身免疫性甲状腺炎的实验研究 被引量:9

Establishment of Experimental Autoimmune Thyroiditis in CBA/J Mice by Different Species and Dosages of Thyroglobulin

摘要 目的探讨利用CBA/J小鼠建立实验性自身免疫性甲状腺炎(EAT)所需甲状腺球蛋白(Tg)的适宜种属和剂量。方法取25只雌性CBA/J小鼠,于8周龄时分别给予猪Tg(pTg)或不同剂量鼠Tg(mTg)加弗氏佐剂进行免疫,或单独给予弗氏佐剂(对照组),10周龄时重复免疫1次,14周龄时处死。利用ELISA法测定鼠血抗Tg抗体(TgAb)水平,并进行甲状腺组织病理检查及炎症评分。结果与对照组相比,CBA/J鼠给予pTg或mTg 100μg免疫后,血TgAb水平均显著升高,甲状腺内有明显淋巴细胞浸润,但给予pTg免疫后滤泡上皮增生较明显。另外,与给予100μg mTg免疫的小鼠相比,给予200μg、400μg mTg免疫小鼠TgAb水平明显更高,淋巴细胞浸润也明显加重,而后2个剂量组相比差异无统计学意义。结论利用CBA/J小鼠建立EAT模型时,mTg为适宜抗原。在本实验条件下,200μg/鼠为最适宜剂量。 Objective To investigate the effects of different species and dosages of thyroglobulin(Tg) on the development of experimental autoimmune thyroiditis(EAT) induced in CBA/J mice.Methods CBA/J female mice were immunized by subcutaneous injection of 100 μg porcine Tg(pTg) or 100 μg,200 μg,400 μg murine Tg(mTg) with Freund’s adjuvant or control vehicle in Freund’s adjuvant(control group) at 8 weeks of age(n =5/group).All of them received a second Tg immunization two weeks later.The mice were euthanized at 14 weeks of age.Serum anti-Tg level was measured using ELISA.Pathological examination and inflammation scoring were performed on paraffin-embedded thyroid tissue sections.Results As compared with the control group,serum TgAb was significantly increased in CBA/J mice immunized with pTg or mTg at the dosage of 100 μg/mouse.Lymphocyte infiltration was present in their thyroid tissues.Hyperplasia of follicular epithelium was observed in the mice immunized with pTg,but not in those with mTg.In addition,serum level of TgAb and the severity of lymphocyte infiltration were significantly increased in the mice immunized with mTg at 200 μg/mouse and 400 μg/mouse compared to those with mTg at 100 μg/mouse,but no significant difference was found between the former two groups.Conclusion mTg is a suitable antigen for the establishment of EAT in the CBA/J mouse.Under our experimental conditions,200 μg mTg per mouse may be the optimum dosage.
