3种复合支架材料用于成骨细胞株体外复合培养的比较

摘要 目的检测细菌纤维素/明胶/羟基磷灰石(BC/GEL/HAP)复合支架材料对成骨细胞附着增殖的影响。方法实验采用小鼠胚胎成骨细胞株(MC3T3-E1)与细菌纤维素(BC)、细菌纤维素/明胶(BC/GEL)、细菌纤维素/明胶/羟基磷灰石(BC/GEL/HAP)3种支架材料进行体外复合培养。通过倒置显微镜观察MC3T3-E1细胞形态,分别于细胞与支架材料复合培养3、6、9 d后采用碱性磷酸酶活性测定法检测成骨细胞与支架材料结合能力,同时扫描电镜观察细胞生长的情况。结果细胞与支架材料复合培养3、6、9 d,实验组与对照组比较,各组之间两两比较,差异均有统计学意义(P<0.05)。结论 3种支架材料均有利于细胞生长,其中细菌纤维素/明胶/羟基磷灰石复合支架材料的增殖效果更好。 Objective To investigate the effect of 3 types of composite scaffold material [bacterial cellulose(BC),BC/gelatin(GEL),and BC/GEL/hydroxyapatite(HAP) on adherence and growth of osteoblasts cultured in vitro.Methods To culture mouse embryonic osteoblasts(cell line MC3T3-E1) in vitro on the 3 types of scaffold materials BC,BC/GEL,and BC/GEL/HAP,and to observe the MC3T3-E1 cell morphology under inverted microscope.After culturing the mouse embryonic osteoblasts(MC3T3-E1)in vitro on the 3 types of scaffold materials for 3,6 and 9 days,respectively,alkaline phosphatase activity was detected to assess the cell adherence to the scaffold material.At the same time,the cell growth was examined by scanning electron microscopy.Results After the embryonic osteoblasts cultured in vitro on the scaffold materials for 3,6,and 9 days,respectively,both the comparison of experimental group with the control group and comparison between each 2 groups showed statistically significant differences(P 0.05).Conclusion All the 3 types of composite scaffold materials BC,BC/GEL and BC/GEL/HAP are good vehicles for adherence and growth of osteoblasts cultured in vitro.Among them,the BC/GEL/HAP composite scaffold material show the best result of osteoblast growth.
