吡格列酮预处理对大鼠脑缺血再灌注损伤保护作用及机制 被引量:3

Mechanism and protective effect of pioglitazone pretreatment on cerebral ischemia-reperfusion injury in rats

摘要 目的研究吡格列酮预处理对缺血再灌注大鼠脑组织的保护作用与机制。方法 75只雄性SD大鼠随机分为假手术组、生理盐水预处理组和吡格列酮预处理组,采用线栓法制作大鼠局灶性脑缺血再灌注模型。缺血2 h再灌注22 h后观察大鼠神经功能评分,测量脑梗死体积,检测脑组织中IL-6、iNOS和NO含量,采用HE染色及Nissl染色观察脑组织病理改变。结果与生理盐水预处理组比较,吡格列酮预处理组大鼠的神经功能评分明显降低、脑梗死体积缩小(P<0.05或P<0.01),脑组织中IL-6、iNOS和NO含量降低(均P<0.01),病理学观察显示其脑组织神经细胞受损程度更小。结论吡格列酮对大鼠局灶性脑缺血再灌注损伤有保护作用,其作用机制与其减轻脑组织炎症反应和NO神经毒性损伤有关。 Objective To investigate the protective effects of pioglitazone pretreatment on focal cerebral ischemia-reperfusion injury in rats and measure the contents of interleukin-6(IL-6) and inducible nitricoxide synthase(iNOS) and nitric oxide(NO) in brain tissue.Methods 75 adult male SD rats were randomly divided into sham operated,normal sodium pretreated and pioglitazone pretreated groups(10 mg/kg).Animal models of 2 h middle cerebral artery occlusion followed by 22 h reperfusion(MCAO/R) were made by suture-occluded method.The neurological score,volume of cerebral infarction were measured.Contents of IL-6,iNOS and NO in brain tissue of rats were investigated.At the same time,the pathological changes of brain tissue were observed by HE staining and Nissl staining.Results Compared with normal sodium group,rats in pioglitazone pretreated group significantly presented lower neurological score and smaller cerebral infarction volume(P0.05),while the contents of IL-6,iNOS and NO in brain tissue were decreased(P0.05).Meanwhile,pathological staining showed smaller injured extent of neurological cells in pioglitazone pretreated group.Conclusions Pioglitazone pretreatment has protective effects on focal cerebral ischemia-reperfusion injury in rats,which is related to the decreased inflammatory reaction and lessened neurotoxicity damage of NO in brain tissue.
