摘要
建立检测金诺芬(AF) 的薄层色谱(TLC) 分析方法。通过对展开剂、吸附剂、显色剂、溶剂、检查方法等的试验, 确定AF 的TLC 分析的最佳条件是氯仿为溶剂, GF254 板作吸附剂, 展开剂是氯仿∶二甲苯∶乙醇∶氨水(60∶30∶9-5∶0-5) , 碘蒸气为显色剂, 检查方法采用自身对照法。在此分析条件下, AF 的Rf 值和灵敏度分别为0-72 ±0-03 , 0-05μg 。AF 中可能存在杂质的Rf 值杂1 为0-65 ±0-03 , 杂2 为0-50 ±0-03 。本法分离效果好、灵敏、简便、结果可靠, 重现性好, 能满足药用AF
A TLC analysis method has been established to detect auranofin(AF).Various of developers,adsorbents,color reagents,solvents and methods of detection were tested.Best results have been obtained by TLC of AF on silica gel GF 254 with the solvent system chloroform-dimethyl benzene-ethyl alcohol-ammonium hydroxide(60∶30∶9 5∶0 5)and chloroform solvent for dissolving AF.Visualizaiton was by exposing the chromatograms to iodine vapor.The dilution of AF was used as reference compound to evaluate AF and related substances.In aforegoing conditions,R f value and sensitivity of AF are 0 72±0 03 and 0 05μg,respectively.In addition,R f values of two impurities which may be contained in the AF are 0 65±0 03 and 0 50±0 03,respectively.This method is successful separation,sensitive,simple and convenient,reliable and reproducible and it is suitable for detecting AF drug.
