摘要
目的和方法:采用膜片钳技术( 细胞贴附式) 研究不同浓度氯化镧(LaCl3) 对培养下丘脑神经元上L- 型钙通道单通道电流的影响。结果:与加药前相比,1 μmol/L 和10 μmol/L LaCl3 分别使L- 型钙通道的开放下降75 % ( P<0-01) ,开放时间缩短44 % ( P< 0-01) ,关闭时间延长53 % ( P< 0-01) ;而0-1 μmol/LLaCl3 分别使L- 型Ca2+ 通道的开放概率增加38 % ( P< 0-05) ,开放时间延长48 % ( P< 0-01) ,关闭时间缩短26 % ( P< 0-05) 。结论:低浓度LaCl3 可兴奋培养下丘脑神经细胞L
AIM and METHODS:Effects of different doses lanthanum chloride (LaCl 3) on L-type calcium channel of cultured hypothalamic neuron were observed by patch clamp recording (cell-attached recording) method.RESULTS:Compared with control before treatment, higher concentration of LaCl 3(1 μmol/L and 10 μmol/L) made a 75% reduction of opening probability ( P <0.01), a 44% shortening of opening time ( P <0.01) and a 53% extension of close time in L-type Ca 2+ channel. However, 0.1 μmol/L LaCl 3 made a 38% increases of opening probability ( P <0.05), a 48% extension of opening time ( P <0.01) and a 26% shortening of close time ( P <0.05) in the L-type Ca 2+ channel compared with control.CONCLUSION:Low LaCl 3 concentration can excite the L-type Ca 2+ channel on culture hypothalamic neurons.
