摘要
为了确定猪繁殖与呼吸综合征病毒(PRRSV)的流行特点及变异规律,本研究首次对从2006年以来我国长江流域分离到的29株PRRSV的ORF3基因进行了克隆和序列测定,并与国外代表毒株和国内的高致病性PRRSV进行了比较分析。结果表明:所有分离毒株的ORF3基因均编码254个氨基酸,都属于美洲型PRRSV,推导的氨基酸同源性为84.6%~100%,与美洲型PRRSV VR-2332的同源性为85.0%~97.2%;与国内分离的高致病性PRRSV的同源性为86.6%~100%。根据ORF3基因的遗传进化分析,这些毒株可分为两个亚群,毒株HZ24与VR-2332及疫苗株亲缘关系较近,其他分离株与高致病性PRRSV处在同一进化分支。同时,该基因编码蛋白的抗原表位和抗原性也发生了变化。
The ORF3 genes of 29 PRRSV,isolated from Changjiang river basin in China from 2006,were amplified,cloned and sequenced with RT-PCR for analyzing the genetic variation of PRRSV,and blast with the representative isolates.The results showed that ORF3 of all isolates encode the minor structural proteins GP3,254 amino acids.All 29 PRRSV isolates belonged to the North American genotype,while 84.6%-100% amino acid identity with each other.It shared 85.0%-97.2% sequence identity with VR-2332,the representative North American strains of PRRSV.And it shared 86.6%-100% sequence identity with the highly pathogenic PRRSV.The phylogenetic trees indicated that the 29 PRRSV were further divided into two subgenotypes,28 isolates posses a same branch with the highly pathogenic PRRSV,and HZ24 shares a more closely relationship with RespPRRS vaccine.Meanwhile,there are mutation in the epitopes and the antigenic index of GP3 in all 29 isolates.
