灵芝多糖对黄颡鱼免疫细胞活性的影响 被引量:26

Effects of Ganoderma lucidum Polysaccharides on the Activation of Immunological Cells of the Yellow Catfish(Pelteobagrus fulvidraco)

摘要 选取540尾黄颡鱼(Pelteobagrus fulvidraco),连续投喂含不同水平灵芝多糖(300,600,900,1 200,1 500mg/kg)的配合饲料8周。而后采用Percoll密度梯度离心等技术,对黄颡鱼头肾巨噬细胞与外周血白细胞进行分离纯化,使用NBT还原法、Griesse试剂显色法与MTT法测定头肾巨噬细胞呼吸爆发活性与外周血白细胞的增殖能力,探讨灵芝多糖对黄颡鱼免疫细胞活性的影响。结果表明,灵芝多糖各水平组均能显著提高黄颡鱼头肾巨噬细胞的氧呼吸爆发活性和外周血白细胞的增殖能力(P<0.01);投喂灵芝多糖饲料后,各组头肾巨噬细胞氮呼吸爆发活性虽有提高,但只有1 200~1 500 mg/kg水平组为极显著提高(P<0.01)。灵芝多糖可以有效提高黄颡鱼免疫细胞的活性。 In this test,540 yellow catfish(Pelteobagrus fulvidraco) were selected,and fed with different levels of Ganoderma lucidum Polysaccharides(300,600,900,1 200,1 500 mg/kg diet,respectively) for 8 weeks.Then the head kidney macrophages and peripheral blood leukocytes of yellow catfish were separated by Percol1 continuous density gradient centrifugation.NBT reduction,Griesse reagent coloration and MTT assay were respectively used to evaluate the respiratory burst activity of head kidney macrophages and proliferation of peripheral blood leukocytes,in order to discuss the effects of GLP on the activation of immunological cells of yellow catfish.The results showed that: the activity of oxygen respiratory burst activity of head kidney macrophages and the activity of proliferation of peripheral blood leukocytes of every levels of GLP group were extremely enhanced(P0.01).Although the nitrogen respiratory burst activities of every level of GLP group were increased,only activities of 1 200-1 500 mg/kg GLP groups were obviously improved(P0.01).As a result,Ganodeima lucidum polysaccharides could effectively enhance the activation of immunological cells of the Yellow catfish.
