摘要
目的 为临床常规检验和进一步研究红细胞补体受体1 型(ECR1) ,建立一种敏感、特异和重复性良好的定量测定方法。方法 戊二醛固定红细胞,并定量“液相包被”于V 型板中,依次加入抗CR1 单克隆抗体、碱性磷酸酶(AP) 标记二抗及可溶性底物显色,移显色液于比色孔,405 nm 比色,计算吸光度值( A) 。结果 正常人ECR1 分子表达数量A 值平均值为1 .26 ±0 .35 ,显著高于肝细胞癌(0 .87 ±0 .21) 和系统性红斑狼疮(0 .42 ±0 .16) 等患者(t= 8 .62 ,P< 0 .01) ,差异有非常显著意义;系统性红斑狼疮患者ECR1 分子表达的数量显著低于肝细胞癌患者(t= 11 .27 ,P< 0 .01) ,差异有非常显著意义,并且ECR1 数量表达与狼疮疾病活动和治疗效果显著相关。结论 本方法具有良好的敏感性、特异性和重复性;CR1 分子的数量表达可分为高、中、低三种形式;肿瘤患者的中、低表达显著升高,狼疮患者无高表达形式。
Objectives To develop a highly sensitive and reproductive test for the number of complement receptor 1 on erythrocytes (ECR1), and to investigate the significance of ECR1 in immune relative diseases.Methods Glutaraldehyde fixed red blood cell (RBC) was analysed in V well microtitre plates. To the cell buttons in each well mouse monoclonal anti CR1 of the first antibody, second antibody of anti mouse IgG conjugated to alkaline phosphatase and the substrate was added following three washes in S/BSA. The supernatant was transferred to a clean U microtitre plate to facilitate reading in a plate reader at 405 nm ( A ).Results ECR1 of the diseases of hepatocellular cancer (0.87±0.21) and systemic lupus erythrematosus (0.42±0.16) was significantly lower than that of healthy individuals (1.26±0.35). The ECR1 of systemic lupus erythrematosus was obviously lower than that of hepatocellular cancer ( t =11.27, P <0.01). Conclusions The technique can examine the number of CR1 on RBC. The number of CR1 can be divided into high, middle and low forms. The middle and low expression of ECR1 was obviously increased in the diseases of cancer, and there is no high expression in the diseases of systemic lupus erythrematosus.
