摘要
目的:建立测定白果中毒性成分总银杏酸含量的方法。方法:采用固相萃取-高效液相色谱法。白果细粉经甲醇回流提取,提取物经C18固相萃取小柱纯化,进样分析。色谱柱为十八烷基硅烷键合硅胶柱(150mm×4.6mm,5μm),流动相为甲醇-3%冰醋酸(91∶9),检测波长为310nm,柱温为30℃,流速为1.0mL·min-1。结果:总银杏酸进样量在0.6219~2.4876μg范围内与峰面积积分值呈良好的线性关系(r=0.9999);平均加样回收率为97.05%,RSD=2.13%(n=6)。结论:本方法简便、准确,适用于白果及其炮制品中总银杏酸的分析,可为白果及其炮制品的质量监控和安全性评价提供研究基础。
OBJECTIVE:To establish the method for the content determination of toxic components ginkgolic acids(GAs) in Ginkgo biloba.METHODS:Solid phase extraction(SPE)-HPLC was adopted.The powder of G.biloba was extracted by methanol and then purified by C18 solid phase extraction column.The determination was performed on C18 column with mobile phase consisted of methanol-3%HAc(91 :9) and detection wavelength of 310 nm.The column temperature was 30 ℃ and flow rate was 1.0 mL·min-1.RESULTS:The linear range of GAs was 0.621 9~2.487 6 μg(r=0.999 9) with an average recovery of 97.05%(RSD=2.13%,n=6).CONCLUSION:The method is simple,accurate and suitable for analyzing GAs in G.biloba and their products.It could provide the basis for the quality control study and safety evaluation of G.biloba and its products.
