动脉粥样硬化血管平滑肌细胞中干细胞因子mRNA的表达水平增加 被引量:1

摘要 目的:分析比较干细胞因子(SCF)、TGF-α及某些膜蛋白、核蛋白基因在不同病理状态下血管平滑肌细胞中的表达水平,探讨这些基因产物在动脉粥样硬化及高血压发病过程中所起的作用。方法:从维生素D3 诱导动脉粥样硬化和一侧肾动脉结扎的高血压大鼠取出主动脉,在体外培养收集血管平滑肌细胞,抽提细胞总RNA,以同位素标记的核酸探针作Northern blot分析。结果:动脉粥样硬化及高血压血管平滑肌细胞中干细胞因子m RNA 表达比正常时高出许多倍,TGFα在高血压血管平滑肌细胞中有较高表达,而erbB2 表达无明显变化。结论:生长因子在血管的损伤修复中起重要作用,实验提示SCF参与了动脉粥样硬化和高血压时动脉平滑肌细胞的增殖调控过程,可能通过干细胞因子这一系统参与血管损伤修复。 Aim:To explore factors influencing on atherosclerotic and hypertension progression,the mRNA expression levels of stem cell factor(SCF),transform growth factor α(TGFα),some membrane proteins and nuclear proteins of vascular smooth muscle cells(VSMCs)in different pathologic conditions were analysed. Methods:Atherosclerosis of rat was induced by vitamin D 3 and hypertension model was prepared by patially obstraction of left renal artery. Total RNA were extracted from cultured vascular smooth muscle cells and analysed with Northern blot. Results:The high level expression of two type of SCF mRNA and incremental TGF α mRNA were found in atherosclerotic and hypertensive vascular smooth muscle cells. Conclusion:Growth factors play an important role in the repair of blood vessel. It is suggested that SCF involved the proliferation control of VSMCs in the atherosclerosis and hypertension by SCF pathway.
