摘要
目的:应用再狭窄动物模型,研究血管成形术后血管平滑肌细胞增殖的分子机制。方法:对家兔实施血管成形术后,分别在术后3、7 和14 天提取实施手术组和对照组的血管总RNA, 应用差异显示技术观察血管成形术后基因表达的改变。结果:我们分别在三个时间点上发现了7 个仅在实施血管成形术的血管表达而在正常对照血管无表达的表达序列标签(Expressed Sequence Tags, ESTs)。尽管这7 个ESTs经序列测定和GenBank 查询均为新的ESTs,但经杂交证实仅有一个EST在损伤血管高表达,而在对照血管表达极低,它可能与血管成形术后血管平滑肌细胞的增殖有关。结论:本研究应用差异显示技术观察了血管成形术后基因表达的改变。利用这一技术可以帮助我们了解疾病动物模型基因表达的变化,这也可能是发现疾病相关基因的一条有效途径。
Aim:To study the molecular mechanisms of vascular smooth muscle cell (VSMC) proliferation after angioplasty in the experimental rabbits. Methods:Balloon angioplasty of the rabbit abdominal artery was performed, and tissues were examined from control animals and from animals, 3 days, 7 days, and 14 days after the operation. Total RNA was isolated, and differential display was performed to identify mRNA transcripts whose expression was modulated in a temporal fashion after balloon angioplasty. Results:Using total RNA isolated from vessels excised from control rabbits and rabbits up to 14 days, we describe seven differentially regulated transcripts by differential display. By cDNA dot blot, only one of the angioplasty induced transcripts at day 7 is highly expressed in vessels from experimental rabbit, and lowly expressed in those from control rabbits, although DNA sequence analysis has identified all of these seven ESTs (Expressed Sequence Tags ) as novel sequences. Conclusion:In this study, we describe in vivo application of the differential display technology in a rabbit abdominal artery angioplasty model to identify mRNA transcripts whose expression is modulated selectively in vessels undergoing neointima. Using models of experimental induced disease should facilitate our understanding of gene expression patterns in pathogenesis and may serve as a reliable technology to find disease related gene.
